Hereditary angioedema is a disease of congenital deficiency or functional defect in the C1 esterase inhibitor (C1-INH) consequent to mutation in theSERPING1gene, which encodes C1-INH. This disease manifests as recurrent, non-pitting, non-pruritic subcutaneous, or submucosal oedema as well as an erythematous rash in some cases. These symptoms result from the uncontrolled localized production of bradykinin.

The most commonly affected sites are the extremities, face, gastrointestinal tract, and respiratory system. When the respiratory system is affected by hereditary angioedema, swelling of the airway can restrict breathing and lead to life-threatening obstruction. As per the National Organization for Rare Disorders (NORD), the prevalence of hereditary angioedema is 1 per 50,000 to 1 in 150,000 individuals worldwide. The expected number of patients in Korea is about 1,000. However, as of 2019, only 80 patients were diagnosed with hereditary angioedema in Korea.

Pharmaceutical drug manufacturers are increasing focus on developing novel therapeutics for hereditary angioedema in order to cater to the large unmet medical needs. Government initiatives and increase in healthcare spending is also fuelling the HAE therapeutics market. For instance, Takeda Pharmaceutical Korea announced that the Ministry of Health and Welfare would grant expanded reimbursement for its acute hereditary angioedema seizure treatment, Firazyr, from March 2021. The ministry will expand the insurance benefits for Firazyr to two doses per patient from the existing one-dose regimen.

Significant number of patients are being misdiagnosed for the condition, also the number of undiagnosed cases it very high which impedes the market growth.
